Imaging Mass Spectrometry

Imaging Mass Spectrometry is a technique that obtains molecular information at each measurement point in a sample together with spatial information to obtain comprehensive molecular map information.

It can be used to analyze a wide variety of sample types, from biological samples to samples made of materials such as metals and films.

Since this method is label-free (no labeling with dyes or isotopes is required), it is less expensive and faster than autoradiography, which is conventionally used to analyze the distribution of compounds, and can analyze the distribution of unchanged compounds and multiple metabolites in tissues.

We perform imaging mass spectrometry using two different techniques called MALDI and DESI.

MALDI

MALDI (matrix assisted laser desorption/ionization) is a method of ionizing sample components by mixing the sample with a matrix (ionization accelerator) and irradiating it with a laser. A wide range of compounds can be analyzed, from low molecular weight compounds to high molecular weight compounds such as proteins.

Because the spot size of the laser irradiation is small and high spatial resolution can be obtained, this method is used for IMS of a wide variety of samples.When performing IMS on tissue sections, it is necessary to apply the matrix solution uniformly to the tissue surface. We can perform IMS by MALDI with the following equipment.

We perform MALDI using the following four types of equipment.

FT-ICR-MS stands for Fourier transform ion cyclotron resonance mass spectrometer, which has the highest mass resolution of any mass spectrometer currently on the market.Because IMS does not use separation methods such as LC/MS, mass spectra are often obtained in the form of mixtures, even if the laser irradiation is localized.FT-ICR-MS, with its ultra-high resolution, allows ultra-selective imaging of the substance of interest. MS/MS is also possible, enabling structural estimation of unknown components.

MALDI-TOF-MS is the most versatile instrument for IMS.It has been used in the Department of Cellular and Molecular Anatomy at Hamamatsu University School of Medicine, and can be used quickly for general-purpose analysis.

TOF-MS with atmospheric pressure MALDI, developed specifically for IMS; like Ultraflex, it is suitable for general-purpose IMS analysis.

This hybrid tandem MS system consists of a quadrupole and a time-of-flight mass separation unit in series, and its high TOF resolution and MS/MS capability make it a powerful tool for the qualitative analysis of unknown components.It is also equipped with a device called ion mobility, which separates ions according to their bulkiness, making it possible to identify isomers that would normally be impossible to identify by MS.Since the analysis is switched with LC/MS, the delivery time may be longer than with other instruments.

DESI

DESI (desorption electrospray ionization) is a method in which electrospray-generated charged droplets are irradiated onto a sample to ionize analytes present on the sample surface.Because it is an electrospray-based ionization method, it is softer than MALDI (less destruction of analytes during ionization) and does not require pretreatment such as matrix application.Spatial resolution is lower than that of MALDI. At our company, IMS by DESI is available for the following instruments.The Department of Cellular and Molecular Anatomy, Hamamatsu University School of Medicine is working on increasing the sensitivity of DESI and applying the results to contract analysis.

This is a triple quadrupole mass spectrometer with quadrupole mass separation sections arranged in series.It is used for quantitative analysis in LC/MS using selected reaction monitoring (SRM). By using this measurement method in DESI/IMS, highly selective IMS can be performed.

Customers can choose the imaging mass spectrometry method that best suits their purpose from two methods, MALDI and DESI.

Through our imaging mass spectrometry business, we provide data to support pharmacokinetic analysis, toxicity prediction, and drug effect analysis of your property compounds.

Liquid chromatography-mass spectrometry (LC/MS)

LC/MS is an instrumental analysis method in which a solution mixture sample is separated by LC to obtain sequential mass spectra.

Compared to other instrumental analysis methods, LC/MS is characterized by high sensitivity and selectivity.

It is also effective for qualitative and quantitative analysis of trace components in complex mixed samples.It is used in various fields such as blood concentration measurement of pharmaceuticals, metabolite analysis of pharmaceuticals, antibody drug analysis, search for disease-related markers, analysis of environmental pollutants, organic material analysis, and food analysis.

In addition to the aforementioned MALDI, electrospray ionization (ESI) for LC/MS is available.Since the system is equipped with a MALDI/ESI dual ion source, switching between both ion sources is instantaneous. A micro HPLC can be connected for LC/MS (/MS) with high sensitivity and ultra-high resolution.

By switching the ion source, MALDI and ESI can be used separately, and the newest LC/MS (/MS) is available by adding a third dimension of separation by ion mobility to component separation by LC and m/z separation by MS.

By switching ion sources, DESI and ESI can be used separately. Mainly using SRM, it is suitable for quantitative analysis of trace constituents containing complex matrices, such as biological samples.

We provide a wide range of LC/MS-related contract analysis services, both qualitative and quantitative, making full use of two types of high-resolution LC-MS/MS and triple quadrupole LC-MS.

Especially for qualitative analysis using high-resolution LC-MS/MS, we can not only measure data but also estimate the structure of unknown compounds.
